introducing Capillogain® Tonic (Topicals & Shampoos)

I would like to introduce to you

Capillogain® Tonic

Capillogain® Tonic contains scientific backed hair growth promoting active agents microencapsulated in so-called Ethosomes. Ethosomes like Liposomes are based on Phosphatidylcholine, however a small fraction of Ethanol causes the Ethosomes to be smaller and their surface gets negatively charged. In addition Ethosomes increase penetration into deeper skin layers over traditional liposomes. However Ethosomes share the time-delayed continuous release of active agents with Liposomes, which promote a prolonged sufficient high level of active agents at the site of the hair follicles with a smooth increase and decrease. It is correct that Ethosomes are also discussed for transdermal drug delivery, however one can formulate Ethosomes also in a way that they deposit the active agents preferably in the skin where they generate a higher skin retention and lower skin permeation which results in less systemic absorption compared to hydro-alcoholic solutions. The rather low alcohol concentration and the complete absence of Propylenglycol (PG) render Capillogain® Tonic more skin-friendly than most commercial hydro-alcoholic products on the market.

A very convincing work on Ethosomes stems from Jun-Bo et al.; the title of their work is:
"Effect of ethosomal minoxidil on dermal delivery and hair cycle of C57BL/6 mice"

Quotes from this work:
"The above results suggested that ethosomes helped drugs reach deeper skin structures, such as pilosebaceous follicles. When the minoxidil was entrapped in the ethosome, 21.94 mg/cm2 minoxidil was delivered into the skin after 12 h. However, less than 8.43 mg/cm2 minoxidil was delivered in the control group where minoxidil is naked."

"After medicine application for 18 days, all of the hairs of ethosomal minoxidil treated group grew out of the mice skin. Histologically, these hair follicles became bigger, and the hair papillae were completely surrounded by the hair bulbs. In the hair bulbs, there were lots of newly formed melanocytes located in the deeper layer of the hypodermis. In addition, newly formed hairs and internal root sheathes were found in most hair bulbs. However, in the control group, the dorsal skins of mice were pink. There were fewer follicles, and less melanocyte formation. The termination of the hair shafts looked like a drumstick. The hair papillae were smaller, and they moved up to the arrectores pilorum attachment level."

"Based on the above animal tests, this observation suggests that ethosomal minoxidil may be a novel strategy for the management of hair growth disorders."

Especially Figure 1 in the publication by Jun-Bo et al. is very interesting since it shows how the active agents were able to reach the dermal papilla with the Ethosomes but nearly no active agents were detected at the dermal papilla with a conventional vehicle.

Currently Capillogain® Tonic is the only topical on the market that contains Procyanidin B-3 from Barley Husk Extract and Raspberry Ketone. Procyanidin B-1, B-2 und C-1 are constituents of Grape Seed Extract and several publications on Procyanidins and hair growth were published in recent years. Procyanidin B-3 turned out to be the most potent Procyanidin and it is a constituent of Barley Husk Extract. The following publications investigated the hair-growth promoting effect of Procyanidin B-3, its anti-inflammatory action, and its antioxidant activity:

“From these results, it is concluded that Procyanidin B-3 can directly promote hair epithelial cell growth in vitro, has the potential to counteract the growth-inhibiting effect caused by TGF-beta1 in vitro, and has potential to stimulate anagen induction in vivo.”
“Topical application of 1% procyanidin oligomers on shaven C3H mice in the telogen phase led to significant hair regeneration [procyanidin B-2, 69.6% +/- 21.8% (mean +/- SD); procyanidin B-3, 80.9% +/- 13.0%; procyanidin C-1, 78.3% +/- 7.6%] on the basis of the shaven area; application of vehicle only led to regeneration of 41.7% (SD = 16.3%). In this paper, we demonstrate the hair-growing activity of procyanidin oligomers both in vitro and in vivo, and their potential for use as agents to induce hair growth.”

“The anti-inflammatory effect of procyanidin B3 (1) on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation of mouse ears was examined. The anti-inflammatory activity of 1 was stronger than that of indomethacin and glycyrrhetinic acid, the normally used anti-inflammatory agents.”

"Although further studies are necessary, these flavan-3-ol extracts show potential to be used to reduce the incidence of degenerative diseases associated with oxidative stress."

“Additionally, ProB3 and RM-1 significantly reduced both the mRNA and protein expression of COX-2 and iNOS in mouse skin tissues (p < 0.01). Such results strongly suggest that ProB3 and RM-1 may be useful in the treatment allergic skin conditions, most notably atopic dermatitis.“

Neutralization of the growth inhibiting effects of TGF-beta is suggested as working mechanism of Procyanidin B-3. TGF-beta is a negative regulator of hair growth as shown in several publications:

The above mentioned inhibition of COX-2 leads to a reduction of Prostaglandin D2 (PGD2), which has recently been shown to be a negative hair growth factor:

Our barley husks stem from Germany and the highly concentrated extract was also produced in Germany.

Hair growth is regulated by positive and negative mediators.
Some of the major positive mediators are:
Keratinocyte Growth Factor 1 (KGF-1, FGF-7), Keratinocyte Growth Factor 2 (KGF-2, FGF-10), Insulin-like Growth Factor 1 (IGF-1), Hepatocyte Growth Factor (HGF), Vascular Endothelial Growth Factor (VEGF), Platelet-Derived Growth Factor AA and BB (PDGF-AA and –BB), and Basic Fibroblast Growth Factor (bFGF, FGF2, FGF-b); list incomplete.

Some of the major negative mediators are:
Fibroblast Growth Factor 5 (FGF-5), Brain-derived Neurotrophic Factor (BDNF), Testosterone (T), Dihydrotestosterone (DHT), Epidermal Growth Factor (EGF), Prostaglandin D2 (PGD2), Transforming Growth Factor beta 1 und 2 (TGF-beta1 and TGF-beta2), Interleukin-6 (IL-6), Dickkopf-1 (DKK-1), 5alpha-Reductase Type 1 and 2 (5AR type 1 and 2), Interferon-gamma (INF-gamma), Interleukin-1beta (IL-1beta), and Androgen Receptor Coactivator Hic-5/ARA55; list incomplete.

It has been shown that the success of Finasteride depends on the IGF-1 level in the scalp:

“In a small uncontrolled study of 9 patients with AGA, an increased expression of IGF-1 messenger RNA levels in the DP was associated with patient response to Finasteride.”

Raspberry Ketone acts on sensory nerve endings similar to Capsaicin. Sensory neurons release calcitonin gene-related peptide (CGRP) on activation by Raspberry Ketone, which in turn increases IGF-1 production in the skin. We hope that the topical application of Raspberry Ketone acts synergistically with oral Finasteride. However, Raspberry Ketone increases IGF-1 in the skin and it already has been shown to be effective even as a stand-alone topical without oral Finasteride in human trials:
„When applied topically to the scalp and facial skin, 0.01% RK promoted hair growth in 50.0% of humans with alopecia (n=10) at 5 months after application and increased cheek skin elasticity at 2 weeks after application in 5 females (p<0.04). These observations strongly suggest that RK might increase dermal IGF-I production through sensory neuron activation, thereby promoting hair growth and increasing skin elasticity.”
All active agents in Capillogain® Tonic are also scientifically backed and chosen to counteract the negative hair growth factors and promote the positive hair growth factors. We tried to cover many different hair growth factors. However, we will soon release a supplemental product Capillogain® Shampoo, which contains additional active agents.

Biotin deficiency leads to pathologic dermatologic changes in the skin and its appendages.

Hinoki Oil
upregulates VEGF expression,
has antibacterial and antifungal effects

gets converted to Phosphatidic Acid which in turn
neutralizes the growth-inhibiting effects of TGF-beta1
and protects the cells from apoptosis

A human trial of Capillogain® Tonic is planned in the near future, however maybe only an experiment in human hair follicle organ culture will be conducted, which is pretty close to human trials.
